Neuroimmmune interactions of cannabinoids in neurogenesis: focus on interleukin-1β (IL-1β) signalling.

چکیده
Neuroimmune networks and the brain endocannabinoid system contribute to the maintenance of neurogenesis. Activation of cannabinoid receptors suppresses chronic inflammatory responses through the attenuation of pro-inflammatory mediators. Moreover, the endocannabinoid system directs cell fate specification of NSCs (neural stem cells) in the CNS (central nervous system). The aim of our work is to understand better the relationship between the endocannabinoid and the IL-1β (interleukin-1β) associated signalling pathways and NSC biology, in order to develop therapeutical strategies on CNS diseases that may facilitate brain repair. NSCs express functional CB1 and CB2 cannabinoid receptors, DAGLα (diacylglycerol lipase α) and the NSC markers SOX-2 and nestin. We have investigated the role of CB1 and CB2 cannabinoid receptors in the control of NSC proliferation and in the release of immunomodulators [IL-1β and IL-1Ra (IL-1 receptor antagonist)] that control NSC fate decisions. Pharmacological blockade of CB1 and/or CB2 cannabinoid receptors abolish or decrease NSC proliferation, indicating a critical role for both CB1 and CB2 receptors in the proliferation of NSC via IL-1 signalling pathways. Thus the endocannabinoid system, which has neuroprotective and immunomodulatory actions mediated by IL-1 signalling cascades in the brain, could assist the process of proliferation and differentiation of embryonic or adult NSCs, and this may be of therapeutic interest in the emerging field of brain repair.

Role of Propolis as a pharmaceutical candidate on interleukin-1β proinflammatory cytokine expression in Alzheimer's rat
Background: One of the important factors that play a key role in the pathogenesis of Alzheimer's disease (AD) is inflammatory processes leading to impaired expression of inflammatory cytokines such as interleukin-6, TNF-α, and interleukin-1β (IL-1β). Propolis, as a therapeutic compound, has anti-inflammatory and antioxidant properties. Therefore, the aim of the present study was to investigate ...
متن کاملSalvianolic acid B improves insulin secretion from interleukin 1β-treated rat pancreatic islets: The role of PI3K-Akt signaling
Background and Objective: Oxidative stress induced by proinflammatory cytokines such as IL-1β plays a major role in β-cell destruction in diabetes type 1. Salvianolic acid B (Sal B) is a polyphenolic compound with antioxidant and protective effects. Thus, objective of this study was to assess the protection exerted by Sal B on isolated rat islets exposed to IL-1β and to investigate an underlyin...

Background: One consequence of diabetes mellitus is increased inflammation, which is exacerbated by training. The objective of this study was to evaluate the effect of eight weeks of aerobic moderate-intensity continuous training on Interleukin-1 beta, Interleukin-13 responses in male Wistar diabetic rats. Methods: 36 diabetic rats were randomly assigned into 3 equal groups (control (C), diabe...
متن کاملAssociation between serum interleukin-1β levels and perinatal asphyxia
Objective: Asphyxia is a major cause of acute mortality and chronic neurologic disability in neonates. We sought to define the predictive values of serum concentrations of interleukin-1β in newborns with perinatal asphyxia to see if there is a relation between interleukin-1β (IL-1β) levels to the short term neurological deficit. Methods: This was a prospective (case-co...

Background: The purpose of this study was to investigate the effect of concurrent training on pre-inflammatory (IL-1β, TNF-α) and anti-inflammatory (IL-10) cytokines gene expression in women with type2 diabetes. Methods: 18-patients (age30-38, >130 glycemic index) were selected, randomly, and divided into control (n=6) the concurrent training(diabetes, n=6) and the concurrent training (healthy...
